BioXcell InVivoMab anti-mouse PD-1 RMP1-14單克隆抗體與小鼠PD-1(也稱為CD279)反應。PD-1是一種50-55kDa的細胞表面受體,由Pdcd1基因編碼,屬于Ig超家族的CD28家族。PD-1在CD4和CD8胸腺細胞以及活化的T和B淋巴細胞和髓細胞上瞬時表達,在成功消除抗原后PD-1的表達下降。此外,在B細胞前階段,Pdcd1 mRNA在發(fā)育中的B淋巴細胞中表達。PD-1的結構包括ITIM(基于免疫受體酪氨酸的抑制基序),這表明PD-1負調控TCR信號。PD-1通過結合B7家族的成員PD-L1和PD-L2發(fā)出信號。在配體結合后,PD-1信號傳導抑制T細胞活化,導致增殖減少,細胞因子產生和T細胞死亡。此外,PD-1敲除動物表現出擴張型心肌病、脾腫大和外周耐受喪失,在小鼠的外周耐受性和預防自身免疫性疾病中發(fā)揮關鍵作用。誘導的PD-L1表達常見于許多腫瘤,包括鱗狀細胞癌、結腸癌和乳腺癌。PD-L1過度表達會導致腫瘤細胞對CD8T細胞介導的裂解的抗性增加。在黑素瘤的小鼠模型中,可以通過用阻斷PD-L1與其受體PD-1之間相互作用的抗體治療來暫時抑制腫瘤生長。由于這些原因,目前正在探索抗PD-1介導的免疫療法作為癌癥治療。與J43抗體一樣,RMP1-14抗體已顯示阻斷小鼠PD-L1-Ig和小鼠PD-L2-Ig與PD-1的結合。
