Enzo Life Sciences的CYTO-ID?綠色/紅色長期細胞示蹤試劑盒采用專有的非共價細胞標記技術(shù),將含有疏水性脂肪鏈的熒光染料穩(wěn)定地結(jié)合入細胞膜的脂質(zhì)雙分子層中。標記緩沖液對哺乳動物細胞是等滲的,不含去污劑或有機溶劑。染料對細胞沒有毒性,可在細胞中保留長達96小時,并在有絲分裂時傳遞給子細胞。由于該染料不對細胞內(nèi)的蛋白質(zhì)進行共價修飾,與基于硫醇反應的氯甲基或胺反應的琥珀酰亞胺酯類熒光染料的分子探針相比,細胞正常的生理反應得到更好的保留。


也可以使用CELLESTIAL?染料進行雙重標記。標記的細胞可以通過共聚焦熒光顯微鏡等儀器進行觀察。此外,染料標記的和未標記的細胞群可通過流式細胞儀進行分析。在延長96小時的培養(yǎng)后,沒有觀察到熒光轉(zhuǎn)移到相鄰細胞。這與Calcein AM和BCECF AM標記形成鮮明對比,后者在生理溫度下只能在存活的細胞內(nèi)保留幾個小時。
